Extracting of the optic disc in optical coherence tomography images

The main target of this work is a design of the program for border localization of the optic disc in OCT data. Part of this program is also localization of the „optic cup“ and calculation of several values that characterize optic disc. The work is also dealing with a description of the optical coherence tomography principle which is used in ophthalmology. One chapter describes some published methods of optic cup localization. Success of the program and its comparison with OCT Zeiss Stratus 3000 is evaluated at the end of the work.
Secretory pathway of plants in pathogene defence

Plants are sessile organisms that have to cope with the changes of their ambient environment. These changes include abiotic disturbances and stresses as well as biotic interactions with other organisms. In many of the biotic interactions, plant cells are hurt or damaged by various bacterial and fungal pathogens. Moreover, plants lack mobile immune cells, which would otherwise render them susceptible to most of these pathogens. Plants have, however, developed other mechanisms for suppressing the infection. Activating the components of the secretory pathway during resistance response is crucial step for suppressing the penetration and growth of pathogens. Fungal pathogens, such as powdery mildews, usually attempt to penetrate the cell wall in order to gain the access to protoplast providing nutrients and to exchange their virulence factors. On the other hand, pathogenic bacteria inhabit extracellular spaces of plant tissues. In both cases, however, the deposition of papilla into the plant cell wall was observed. Papilla thus seems to be the common defence mechanism. There is an increasing evidence confirming the indispensable role of polarized secretion in formation of papilla. My PhD thesis has addressed specific questions regarding cell proliferation and tissue dynamics in three key areas of craniofacial development: during suppression and revitalization of tooth buds that develop in the mouse embryonic dentition as rudiments of lost premolars; during dental papilla and follicle formation of the first mouse molar; and during origin of palatal rugae on the mouse hard palate. By evaluation of cell proliferation, we recorded a change in the proliferation pattern along the cheek region of the mandible between less and more advanced embryos at embryonic day 13.5. Thus during the time period, when the development of the large mouse rudimental premolar primordium (R2) is stopped, we showed that the arrest of the rudiment R2 is caused by exhibiting low rate of cell proliferation and high rate of apoptosis. When Sprouty gene signalling is disrupted, the premolar primordium shows rates of proliferation and apoptosis similar to the growing first molar. The R2 subsequently revitalizes and develops into a supernumerary tooth in front of the first molar.
